چکیده

The antagonism, by neostigmine, of neuromuscular blockade produced by either Org NC 45 or pancuronium was studied in 29 anaesthetized patients during a continuous infusion of the myoneural blocker. The ED50 of neostigmine (dose which produced a 50% antagonism) when antagonizing Org NC 45 and pancuronium was 0.011 mg kg-1 and 0.010 mg kg-1 respectively. The dose-response relationships for the antagonism of Org NC 45 and pancuronium neuromuscular blockades were not significantly different. The duration of the effect of neostigmine was not different when antagonizing an Org NC 45- or pancuronium-induced blockade. We concluded that Org NC 45 and pancuronium are effectively and equally (independent of pharmacokinetics) antagonized by neostigmine in man.

Clinical studies on Org NC 45: comparison with pancuronium.

Some actions of Org NC 45 have been compared with those of pancuronium in a study on 50 patients anesthetized with nitrous oxide and narcotic supplements. The train-of-four technique was used to assess the recovery of neuromuscular blockade and its antagonism. Org NC 45 was shown to have a duration of action from injection of the intubating dose (0.1 mg kg-1) to the reappearance of the first tw...

Comparison of the neuromuscular blocking properties of ORG NC 45 and pancuronium in the rat, cat and rhesus monkey.

The neuromuscular and cardiovascular activities of pancuronium were compared with those of its 16(3 monoquaternary congener, Org NC 45, in the anaesthetized rat, cat and rhesus monkey. In the rat Org NC 45 was 3.4 times less potent as a neuromuscular blocking agent than was pancuronium. In the cat, both drugs were found to be approximately equally eliminated by the liver, whereas renal eliminat...

Omitting antagonism of neuromuscular block: effect on postoperative nausea and vomiting and risk of residual paralysis. A systematic review.

We have estimated the effect of omitting antagonism of neuromuscular block on postoperative nausea and vomiting. A systematic search (MEDLINE, EMBASE, Biological Abstracts, Cochrane library, reference lists and hand searching; no language restriction, up to March 1998) was performed for relevant randomized controlled trials. In eight studies (1134 patients), antagonism with neostigmine or edrop...

Atracurium, vecuronium and pancuronium in end-stage renal failure. Dose-response properties and interactions with azathioprine.

Dose-response relations for atracurium, vecuronium and pancuronium were determined in patients in end-stage renal failure for the initial neuromuscular blockade (using three cumulative doses) and for the maintenance of stable 90% response (during continuous infusion). All measurements were during renal transplant surgery, and the interaction of azathioprine on neuromuscular blockade was estimat...

Is recovery of neuromuscular transmission complete after the use of neostigmine to antagonize block produced by rocuronium, vecuronium, atracurium and pancuronium?

To test if recovery of neuromuscular transmission is complete after the use of neostigmine under standardized conditions, we have measured adductor pollicis mechanical activity in response to 0.1 Hz (twitch height), train-of-four (TOF) and 100 Hz (RF 100 Hz) ulnar nerve stimulations. We studied 56 adult anaesthetized (thiopentone, fentanyl, nitrous oxide in oxygen) patients, allocated randomly ...
